چکیده

latter were present, the amount of label in the glycerol moiety of the triacylglycerols was increased &fold and that in the diacylglycerol fraction was increased 3-fold. Fatty acids derived from glucose were found mainly in the triacylglycerols, especially in the presence of exogenous free fatty acids, although appreciable amounts of label again accumulated in the diacylglycerols. The finding that fatty acids, formed de nouo from acetate or glucose, could accumulate in the diacylglycerols in rat adipose tissue appeared to be at variance with the observation that the hydrolysis of phosphatidic acid to diacylglycerol by phosphatidate phosphohydrolase is the rate-limiting step in triacylglycerol biosynthesis in rat adipose tissue (Christie et al., 1976). Fatty acid synthesis de nouo may also have a controlling function.

Studies on the biosynthesis of protein by lactating guinea-pig mammary gland. Characteristics of the synthesis of alpha-lactalbumin and total protein by slices and cell-free systems.

1. Slices of lactating guinea-pig mammary gland were incubated with radioactive amino acids and the various subcellular fractions separated by centrifugation after disruption of the cells by mincing and homogenization. The most active fraction for protein synthesis appeared to be the ;mitochondrial'. 2. When the subcellular fractions were prepared without previous incubation of the cells and we...

Chain-Termination Mechanism in Fatty Acid Synthesis by Mammalian Fatty Acid Synthetase SYNTHESIS OF BUTYRATE AND HEXANOATE BY LACTATING COW MAMMARY GLAND

1. Purified cow mammary gland fatty acid synthetase synthesized long-chain unesterifled and short-chain esterified fatty acids. 2. A direct relationship was observed between the amount of short-chain products synthesized and the concentration of acetyl-CoA in the incubation medium. 3. The short-chain products were identified as butyryl-CoA and hexanoyl-CoA. 4. Inhibition of the terminating thio...

Epoxide-metabolizing enzymes in mammary gland and liver from BALB/c mice and effects of inducers on enzyme activity.

Epoxide hydrolases (EC 3.3.2.3) (EH) are hydrolytic enzymes which may play an important role in the activation and detoxification of mammary carcinogens. In this study, microsomal, cytosolic, and cholesterol epoxide hydrolases along with glutathione S-transferase were characterized in liver and mammary gland from nulliparous and lactating BALB/c mice and from mice transplanted with preneoplasti...

Acyl specificity in triglyceride synthesis by lactating rat mammary gland.

We have studied the specificity of the acyl-CoA:diglyceride acyltransferase reaction in lactating rat mammary gland to provide a rational explanation at the enzyme level for the nonrandom distribution of fatty acids in milk fat triglycerides. Acyl-CoA:diglyceride acyltransferase activity was measured using various diglyceride and radioactive acyl-CoA substrates; products were identified as trig...

Lipid-linked oligosaccharides containing glucose in lactating rabbit mammary gland.

1. Microsomal fractions of lactating rabbit mammary gland incubated with UDP-glucose formed lipid-linked mono- and oligo-saccharides. The lipid-linked monosaccharide had chromatographic properties similar to those of dolichol phosphate mannose and yielded glucose on acid hydrolysis. 2. Incubation of the microsomal fraction with GDP-[U14C]-mannose yielded an oligosaccharide lipid of approximatel...
